PT-141: For Better or Worse

Plus my guide on how to use it

There’s no question that PT-141 (Bremelanotide) is one of the most intriguing peptides available.

My comprehensive video on it is the second highest viewed video on my channel and currently has 50k+ views.

We recently began carrying PT-141 at BioLongevity Labs.

Today, I will walk through how it can potentially be a great alternative (or addition) to using drugs like Viagara or Cialis and how men AND women can both use it.

Let’s dive in.

What Does the Science Say About PT-141?

First things first—PT-141 works differently than drugs like Viagra or Cialis.

Instead of just increasing blood flow, it acts directly on the nervous system, stimulating the melanocortin receptors responsible for sexual arousal.

And guess what? Unlike some other peptides, we’ve got the numbers to prove it.

  • Increased Sexual Desire in Women

    • Two Phase 3 clinical trials showed that premenopausal women with Hypoactive Sexual Desire Disorder (HSDD) had a statistically significant increase in sexual desire when using PT-141. (Source)

  • Faster Onset of Action

    • Unlike oral medications, PT-141 starts working within 30 to 60 minutes after subcutaneous injection. (Source)

  • Lasts Up to 16 Hours

    • The effects of PT-141 don’t just vanish after a couple of hours. They can last up to 16 hours, making it far more versatile than other sexual enhancement options. (Source)

  • Improvement in Erectile Function

    • In a clinical trial on men, doses of 10mg and 20mg significantly improved the duration of erections, with base rigidity ≥80% compared to placebo. (Source) WARNING: DO NOT START WITH THIS HIGH OF A DOSE (I’ll explain further below).

  • Common Side Effects

    • Like any powerful peptide, PT-141 isn’t without side effects. There is a certain subsection of users who do not get pleasant effects. The most commonly reported are:

      • Nausea (40%)

      • Flushing (20%)

      • Headaches (11%)

      • (Source)

  • Blood Pressure Considerations

    • You should be aware that PT-141 can cause a temporary spike in blood pressure (~6 mm Hg systolic, 3 mm Hg diastolic) that returns to normal after 12 hours. (Source)

  • Patient Satisfaction Rates

    • Women using PT-141 reported higher satisfaction levels on the Patient Global Impression of Improvement scale compared to placebo. (Source)

  • Improvement in Satisfying Sexual Events (SSEs):

    • A pooled analysis of doses (1.25/1.75 mg) of Bremelanotide demonstrated a statistically and clinically significant increase in the number of SSEs per month, which was the primary outcome measure of the study. (Source)

  • Safety and tolerability:

    • In clinical evaluations, PT-141 was safely administered and well-tolerated, with no maximum-tolerated dose identified. Common side effects included flushing and nausea. (Source)

So, Is PT-141 Worth It?

Yes, IF you don’t get nausea or a flushing reaction.

I personally LOVE using it, while my business partner Jay Campbell gets debilitating nausea after injecting.

Let’s break it down:

Yes, it works. The data is clear—PT-141 increases sexual desire and arousal in both men and women.

Yes, it works differently. Unlike PDE5 inhibitors, it works on brain chemistry, not just blood flow.

Yes, it has side effects. Nausea, flushing, and headaches are common, so don’t expect a free ride.

But if you’re looking for something beyond the typical “blue pill” and want to enhance both desire and performance, PT-141 might be one of the most powerful tools.

Don’t forget my favorite caveat!

Just like with GLP-1 peptides, people think PT-141 will do all the work for them.

Think you can inject and suddenly transform into a sexual powerhouse overnight?

Nope.

Doesn’t work like that.

You STILL MUST:

Optimize your hormones – If your testosterone, estrogen, or dopamine levels are off, PT-141 won’t work as well.

Dial in your lifestyle – If you’re sleep-deprived, sedentary, or eating garbage, no peptide will fix that.

Use it correctly – It’s not a “take it and immediately perform” drug. You need to time it properly and use the right stack for best results.

So, if you’re considering PT-141 (or are already using it), make sure you understand what you’re doing—or risk wasting your time and money.

Use Guide

If you’re ready to use PT-141 correctly, here’s exactly how to mix, dose, and inject it for optimal results.

Step 1: Reconstituting PT-141

Your PT-141 vial likely comes in a 10mg powder form. You’ll need to reconstitute it with bacteriostatic water before use.

  1. Use 2ml of bacteriostatic water to mix into your 10mg vial.

  2. Gently swirl (don’t shake) until the powder is fully dissolved.

Step 2: Dosing PT-141 Correctly

Now that your vial is mixed, here’s the math:

  1. Every 0.1ml (10 units on an insulin syringe) = 0.5mg of PT-141

  2. A standard starting dose is 1mg (0.2ml or 20 units on the syringe)

Common dosing protocol:

Begin with 1mg (0.2ml/20 units) as a subcutaneous injection.

Increase to 1.5mg-2mg if needed, based on response.

Administer 1-4 hours before desired effects.

Avoid exceeding 2mg per dose to minimize nausea and side effects.

Step 3: Timing & Frequency

  • PT-141 works best when injected 1-4 hours before activity (effects peak between 2-6 hours).

  • For most users, once or twice per week is sufficient.

  • Effects can last up to 16 hours, but individual responses vary.

Step 4: Managing Side Effects

  • Nausea is the #1 issue. If you experience nausea, try lowering your dose or splitting it into two smaller injections.

  • Stay hydrated and eat something light if you feel off.

Step 5: Storing PT-141

  • Store unreconstituted PT-141 in the fridge or a cool, dark place.

  • Once mixed, keep it refrigerated and use it within 90 days for best potency.
